METHOD 
METHOD Comparative modeling was used to predict the structure of T0073.  A 
METHOD database search identified several potential template structures,  
METHOD including 1cos, which was selected as template.  Secondary structure 
METHOD prediction (phd) suggested a turn in the sequence at approximately  
METHOD residues GFSP.  A description of the 1cos structure (Lovejoy et al., 1993) 
METHOD discusses the characteristic heptad repeats designed in this molecule, 
METHOD as well as the leucine stacking within the interior of the three-helix  
METHOD bundle.  The characteristic heptad repeats of 1cos are also found in  
METHOD the sequence of T0073.  We predict that a homodimer of T0073 folds 
METHOD to form a three-helix bundle, similar to 1cos.  In 1cos, two of the three 
METHOD helices are parallel to each other (Helices I and II) and one is  
METHOD antiparallel to those two (Helix III).  We chose model each monomer  
METHOD as a helix with a turn at residues GFSPEE, with a short helical segment 
METHOD at the N-terminus and a longer helical segment at the C-terminus.  We 
METHOD assume each monomer will take on the approximate conformation of a  
METHOD candycane, so that two monomers will form a three-helix bundle.  We 
METHOD predict the long (C-termini) helices of the two monomers will resemble  
METHOD helices I and III in the 1cos structure.  We predict the short (N-terminal)  
METHOD helices will fold together to form the third helix of the three-helix 
METHOD bundle, analogous to helix II of 1cos, with the N-termini in the middle 
METHOD of the structure and the helices going in opposite directions.  Time  
METHOD constraints prohibited us from properly modelling the loop (GFSPEE) 
METHOD so this region was omitted.

METHOD NOTE:  WE BELIEVE THIS MOLECULE IS A HOMODIMER, BUT HAVE SUBMITTED 
METHOD THE TWO MONOMERS AS MODEL1 AND MODEL2 BECAUSE THERE IS NO OTHER 
METHOD FOR US TO SUBMIT IT WITH THE GIVEN FORM.  WE BELIEVE BOTH MONOMERS 
METHOD OF THE MODEL ARE NECESSARY FOR A PROPER EVALUATION.
